Background and purpose: Periventricular white matter hyperintensities (WMH; PVWMH) and deep WMH (DWMH) are regional classifications of WMH and reflect proposed differences in cause. In the first study, to date, we undertook genome-wide association analyses of DWMH and PVWMH to show that these phenotypes have different genetic underpinnings. Methods: Participants were aged 45 years and older, free of stroke and dementia. We conducted genome-wide association analyses of PVWMH and DWMH in 26,654 pa... Abstract

Expert review of cardiovascular therapy, Volume 18, Issue 6, 9 2 2020, Pages 363-372 Anticoagulant treatment and bleeding complications in patients with left ventricular assist devices. den Exter PL, Beeres SLMA, Eikenboom J, Klok FA, Huisman MV
Introduction: The treatment options for advanced heart failure patients drastically changed with the introduction of left ventricular assist devices (LVADs), either as bridge to transplant or as destination therapy for patients ineligible for transplant. Despite major benefits in terms of survival, functional status and quality of life, managing patients with LVADs comes with several challenges. The most significant challenge is balancing between the risks of thrombotic and bleeding complication... Abstract

eLife, Volume 9, 8 2 2020, Pages e55921 Self-restoration of cardiac excitation rhythm by anti-arrhythmic ion channel gating. Majumder R, De Coster T, Kudryashova N, Verkerk AO, Kazbanov IV, Ördög B, Harlaar N, Wilders R, de Vries AA, Ypey DL, Panfilov AV, Pijnappels DA
Homeostatic regulation protects organisms against hazardous physiological changes. However, such regulation is limited in certain organs and associated biological processes. For example, the heart fails to self-restore its normal electrical activity once disturbed, as with sustained arrhythmias. Here we present proof-of-concept of a biological self-restoring system that allows automatic detection and correction of such abnormal excitation rhythms. For the heart, its realization involves the inte... Abstract

Histopathology, Volume 77, Issue 1, 7 1 2020, Pages 92-99 Performance of the pattern-based interpretation of p53 immunohistochemistry as a surrogate for TP53 mutations in vulvar squamous cell carcinoma. Kortekaas KE, Solleveld-Westerink N, Tessier-Cloutier B, Rutten TA, Poelgeest MIE, Gilks CB, Hoang LN, Bosse T
Aims: The most commonly mutated gene in vulvar squamous cell carcinoma (VSCC) is TP53 and its prognostic value, particularly in HPV-independent VSCC, is uncertain. In other tumours, p53 immunohistochemistry (IHC) is an excellent surrogate marker for TP53 mutations. In order to study this in VSCC, we assigned six p53 IHC patterns into two final classes: 'wild-type' or 'mutant'. We determined the performance and interobserver variability of this pattern-based p53 IHC approach.... Abstract

Nederlands tijdschrift voor geneeskunde, Volume 164, 4 1 2020, Pages D4191 [Pharmacogenetics in daily practice]. van Gelder T, van Schaik RHN
With the exception of a few medical specialties, the implementation of pharmacogenetic tests in daily practice has thus far been limited. The Royal Dutch Pharmacists Association (KNMP) has developed pharmacogenetics-based therapeutic doserecommendations for 80 medicinal product combinations on the basis of a systematic literature review. Genotyping of patients can take place on a reactive or pre-emptive basis; the advantage of pre-emptive genotyping is that it provides genetic information the mo... Abstract

Bioinformatics (Oxford, England), Volume 36, Issue 12, 1 1 2020, Pages 3874-3876 gplas: a comprehensive tool for plasmid analysis using short-read graphs. Arredondo-Alonso S, Bootsma M, Hein Y, Rogers MRC, Corander J, Willems RJL, Schürch AC
Summary: Plasmids can horizontally transmit genetic traits, enabling rapid bacterial adaptation to new environments and hosts. Short-read whole-genome sequencing data are often applied to large-scale bacterial comparative genomics projects but the reconstruction of plasmids from these data is facing severe limitations, such as the inability to distinguish plasmids from each other in a bacterial genome. We developed gplas, a new approach to reliably separate plasmid contigs into discrete componen... Abstract

Human nature (Hawthorne, N.Y.), Volume 31, Issue 2, 1 1 2020, Pages 155-173 Why Class Formation Occurs in Humans but Not among Other Primates : A Primate Coalitions Model. Pandit SA, Pradhan GR, van Schaik CP
Most human societies exhibit a distinct class structure, with an elite, middle classes, and a bottom class, whereas animals form simple dominance hierarchies in which individuals with higher fighting ability do not appear to form coalitions to "oppress" weaker individuals. Here, we extend our model of primate coalitions and find that a division into a bottom class and an upper class is inevitable whenever fitness-enhancing resources, such as food or real estate, are exploitable or trad...
