EMBO reports, Volume 15, Issue 1, 15 3 2013, Pages 62-69 Biased competition between Lgr5 intestinal stem cells driven by oncogenic mutation induces clonal expansion. Snippert HJ, Schepers AG, van Es JH, Simons BD, Clevers H
The concept of 'field cancerization' describes the clonal expansion of genetically altered, but morphologically normal cells that predisposes a tissue to cancer development. Here, we demonstrate that biased stem cell competition in the mouse small intestine can initiate the expansion of such clones. We quantitatively analyze how the activation of oncogenic K-ras in individual Lgr5(+) stem cells accelerates their cell division rate and creates a biased drift towards crypt clonality. K-r... Journal of biomolecular screening, Volume 19, Issue 2, 11 2 2013, Pages 287-296 A novel microscopy-based high-throughput screening method to identify proteins that regulate global histone modification levels. Baas R, Lelieveld D, van Teeffelen H, Lijnzaad P, Castelijns B, van Schaik FM, Vermeulen M, Egan DA, Timmers HT, de Graaf P
Posttranslational modifications of histones play an important role in the regulation of gene expression and chromatin structure in eukaryotes. The balance between chromatin factors depositing (writers) and removing (erasers) histone marks regulates the steady-state levels of chromatin modifications. Here we describe a novel microscopy-based screening method to identify proteins that regulate histone modification levels in a high-throughput fashion. We named our method CROSS, for Chromatin Regula... Abstract

Neural computation, Volume 26, Issue 3, 9 2 2013, Pages 472-496 Approximate, computationally efficient online learning in Bayesian spiking neurons. Kuhlmann L, Hauser-Raspe M, Manton JH, Grayden DB, Tapson J, van Schaik A
Bayesian spiking neurons (BSNs) provide a probabilistic interpretation of how neurons perform inference and learning. Online learning in BSNs typically involves parameter estimation based on maximum-likelihood expectation-maximization (ML-EM) which is computationally slow and limits the potential of studying networks of BSNs. An online learning algorithm, fast learning (FL), is presented that is more computationally efficient than the benchmark ML-EM for a fixed number of time steps as the numbe... Abstract

BMC nephrology, Volume 14, 9 2 2013, Pages 273 Time of injury affects urinary biomarker predictive values for acute kidney injury in critically ill, non-septic patients. de Geus HR, Fortrie G, Betjes MG, van Schaik RH, Groeneveld AB
Background: The predictive value of acute kidney injury (AKI) urinary biomarkers may depend on the time interval following tubular injury, thereby explaining in part the heterogeneous performance of these markers that has been reported in the literature. We studied the influence of timing on the predictive values of tubular proteins, measured before the rise of serum creatinine (SCr) in critically ill, non-septic patients. Methods: Seven hundred adult critically ill patients were prospectively i... Abstract
