Biochemical pharmacology, Volume 68, Issue 11, 1 1 2004, Pages 2273-2281 A methylester of the glucuronide prodrug DOX-GA3 for improvement of tumor-selective chemotherapy. de Graaf M, Nevalainen TJ, Scheeren HW, Pinedo HM, Haisma HJ, Boven E
The glucuronide prodrug of doxorubicin, DOX-GA3, can be selectively activated in tumors by extracellular human beta-glucuronidase, resulting in a better therapeutic index than doxorubicin. DOX-GA3, however, is rapidly excreted by the kidney. We hypothesized that slow release of DOX-GA3 from its methylester, DOX-mGA3, by esterase activity in blood would result in improved circulation half-life (t(1/2)) of DOX-GA3. DOX-mGA3 was synthesized more efficiently with an overall yield of 60% as compared... Abstract

Clinical and experimental allergy : journal of the British Society for Allergy and Clinical Immunology, Volume 34, Issue 12, 1 1 2004, Pages 1827-1833 Serum surfactant protein D is elevated in allergic patients. Koopmans JG, van der Zee JS, Krop EJ, Lopuhaä CE, Jansen HM, Batenburg JJ
Background: There is evidence that surfactant protein (SP)-D is important in the innate, as well as in the adaptive pulmonary immune response. Serum concentrations of SP-D have been proposed as parameter of the integrity of the blood-airspace barrier in interstitial lung diseases. We hypothesized that serum SP-D concentrations are affected in allergic patients and correlate with changes in allergic airway inflammation. Objective: To determine levels of serum SP-D in allergic patients compared wi... Abstract

Inorganic chemistry, Volume 43, Issue 26, 1 1 2004, Pages 8461-8469 Rare-earth quinolinates: infrared-emitting molecular materials with a rich structural chemistry. Van Deun R, Fias P, Nockemann P, Schepers A, Parac-Vogt TN, Van Hecke K, Van Meervelt L, Binnemans K
Near-infrared-emitting rare-earth chelates based on 8-hydroxyquinoline have appeared frequently in recent literature, because they are promising candidates for active components in near-infrared-luminescent optical devices, such as optical amplifiers, organic light-emitting diodes, .... Unfortunately, the absence of a full structural investigation of these rare-earth quinolinates is hampering the further development of rare-earth quinolinate based materials, because the luminescence output canno... Gene, Volume 343, Issue 1, 1 1 2004, Pages 91-97 Efficient somatic gene targeting in the lymphoid human cell line DG75. Feederle R, Delecluse HJ, Rouault JP, Schepers A, Hammerschmidt W
Among the different approaches used to define the function of a protein of interest, alteration and/or deletion of its encoding gene is the most direct strategy. Homologous recombination between the chromosomal gene locus and an appropriately designed targeting vector results in an alteration or knockout of the gene of interest. Homologous recombination is easily performed in yeast or in murine embryonic stem cells, but is cumbersome in more differentiated and diploid somatic cell lines. Here we... Abstract

Pacing and clinical electrophysiology : PACE, Volume 27, Issue 12, 1 1 2004, Pages 1603-1609 Hypertensive stress increases dispersion of repolarization. Van Huysduynen BH, Swenne CA, Van Eck HJ, Kors JA, Schoneveld AL, Van De Vooren H, Schiereck P, Schalij MJ, Van Der Wall EE
Several electrocardiographic indices for repolarization heterogeneity have been proposed previously. The behavior of these indices under two different stressors at the same heart rate (i.e., normotensive gravitational stress, and hypertensive isometric stress) was studied. ECG and blood pressure were recorded in 56 healthy men during rest (sitting with horizontal legs), hypertensive stress (performing handgrip), and normotensive stress (sitting with lowered legs). During both stressors, heart ra... Abstract

Circulation, Volume 110, Issue 20, 8 2 2004, Pages 3187-3192 Left atrial tachycardia originating from the mitral annulus-aorta junction. Gonzalez MD, Contreras LJ, Jongbloed MR, Rivera J, Donahue TP, Curtis AB, Bailey MS, Conti JB, Fishman GI, Schalij MJ, Gittenberger-de Groot AC
Background: At the mitral annulus-aorta (MA-Ao) junction, the left atrium is continuous through the subaortic curtain with the musculature of the anterior mitral leaflet. Under experimental conditions, this region can generate abnormal electrical activity. In patients with left atrial tachycardia, we investigated whether this region could be the source of this arrhythmia. Methods and results: In 10 (28%) of 35 consecutive patients with left atrial tachycardia, the arrhythmia originated from the... Abstract

Pediatric research, Volume 57, Issue 1, 5 1 2004, Pages 16-21 Systolic and diastolic ventricular function assessed by pressure-volume loops in the stage 21 venous clipped chick embryo. Stekelenburg-de Vos S, Steendijk P, Ursem NT, Wladimiroff JW, Delfos R, Poelmann RE
Cardiac pressure-volume relations enable quantification of intrinsic ventricular diastolic and systolic properties independent of loading conditions. The use of pressure-volume loop analysis in early stages of development could contribute to a better understanding of the relationship between hemodynamics and cardiac morphogenesis. The venous clip model is an intervention model for the chick embryo in which permanent obstruction of the right lateral vitelline vein temporarily reduces the mechanic... Abstract

Journal of the American College of Cardiology, Volume 44, Issue 9, 1 1 2004, Pages 1834-1840 Left ventricular dyssynchrony predicts response and prognosis after cardiac resynchronization therapy. Bax JJ, Bleeker GB, Marwick TH, Molhoek SG, Boersma E, Steendijk P, van der Wall EE, Schalij MJ
Objectives: This study was designed to predict the response and prognosis after cardiac resynchronization therapy (CRT) in patients with end-stage heart failure (HF). Background: Cardiac resynchronization therapy improves HF symptoms, exercise capacity, and left ventricular (LV) function. Because not all patients respond, preimplantation identification of responders is needed. In the present study, response to CRT was predicted by the presence of LV dyssynchrony assessed by tissue Doppler imagin... Abstract

The Journal of clinical endocrinology and metabolism, Volume 89, Issue 11, 1 1 2004, Pages 5828-5836 Involvement of membrane-type matrix metalloproteinases (MT-MMPs) in capillary tube formation by human endometrial microvascular endothelial cells: role of MT3-MMP. Plaisier M, Kapiteijn K, Koolwijk P, Fijten C, Hanemaaijer R, Grimbergen JM, Mulder-Stapel A, Quax PH, Helmerhorst FM, van Hinsbergh VW
In the endometrium, angiogenesis is a physiological process, whereas in most adult tissues neovascularization is initiated only during tissue repair or pathological conditions. Pericellular proteolysis plays an important role in angiogenesis being required for endothelial cell migration, invasion, and tube formation. We studied the expression of proteases by human endometrial microvascular endothelial cells (hEMVECs) and their involvement in the formation of capillary tubes and compared these re... Abstract

Echocardiography (Mount Kisco, N.Y.), Volume 21, Issue 8, 1 1 2004, Pages 721-723 Transesophageal echocardiography-guided transvenous biopsy of an intracardiac tumor. Scholte AJ, Frissen PH, van der Wouw PA
Transesophageal echocardiography with simultaneous fluoroscopic guidance during a transvenous biopsy of an intracardiac tumor is a valuable technique. We present the case of a 67-year-old man with a metastasis in the right atrium from which histological examination of the tumor was indicated. The inaccessible location of the primary tumor in the liver and impaired hemostasis due to cirrhosis necessitated a transvenous biopsy of the metastasis. The procedure and the associated techniques are disc... Abstract

Circulation, Volume 110, Issue 19, 1 1 2004, Pages 3017-3022 Abnormal myocardial presynaptic norepinephrine recycling in patients with Brugada syndrome. Kies P, Wichter T, Schäfers M, Paul M, Schäfers KP, Eckardt L, Stegger L, Schulze-Bahr E, Rimoldi O, Breithardt G, Schober O, Camici PG
Background: Life-threatening ventricular tachyarrhythmias can occur in young patients without structural heart disease (idiopathic forms). In many patients, these are typically triggered by an increased sympathetic tone, eg, by physical or mental stress. In contrast, in Brugada syndrome, ventricular tachyarrhythmias more often occur during rest or sleep when the vagal tone is predominant. Furthermore, adrenergic agonists can reduce the level of ST-segment elevation, whereas it is increased by pa... Abstract
