Atherosclerosis, Volume 162, Issue 1, 1 1 2002, Pages 103-110 VLDL receptor deficiency enhances intimal thickening after vascular injury but does not affect atherosclerotic lesion area. Tacken PJ, Delsing DJ, Gijbels MJ, Quax PH, Havekes LM, Hofker MH, van Dijk KW
The very low density lipoprotein receptor (VLDLR) has been shown to modulate cell migration and foam cell formation in vitro. This suggests a role for the VLDLR in vascular pathology associated with intimal thickening and atherogenesis. In the present paper both intimal thickening and atherosclerosis were studied using VLDLR knockout and transgenic mouse models. The role of the VLDLR in intimal thickening was established in an in vivo model for vascular injury. A non-restrictive cuff was placed... Abstract

Circulation research, Volume 90, Issue 10, 1 1 2002, Pages 1128-1134 Adenoviral activin a expression prevents intimal hyperplasia in human and murine blood vessels by maintaining the contractile smooth muscle cell phenotype. Engelse MA, Lardenoye JH, Neele JM, Grimbergen JM, De Vries MR, Lamfers ML, Pannekoek H, Quax PH, De Vries CJ
Activin A alters the characteristics of human arterial smooth muscle cells (SMCs) toward a contractile, quiescent phenotype. We hypothesize that activin A may prevent SMC-rich neointimal hyperplasia. Here, we study the effect of adenovirus-mediated expression of activin A on neointima formation in vitro and in vivo. Human saphenous vein organ cultures, in which a neointima is formed spontaneously, were infected either with activin A- or lacZ-adenovirus. Activin A-overexpression reduces neointima... Abstract

European journal of nuclear medicine and molecular imaging, Volume 29, Issue 6, 10 2 2002, Pages 721-727 Accuracy of PET in predicting functional recovery after revascularisation in patients with chronic ischaemic dysfunction: head-to-head comparison between blood flow, glucose utilisation and water-perfusable tissue fraction. Bax JJ, Fath-Ordoubadi F, Boersma E, Wijns W, Camici PG
Accurate prediction of improvement in left ventricular ejection fraction (LVEF) after revascularisation is critical in the therapeutic decision-making process in patients with chronic ischaemic dysfunction. Positron emission tomography (PET) allows non-invasive evaluation of myocardial blood flow (MBF), metabolic rate of glucose (MRG, absolute and relative) and the water-perfusable tissue fraction (PTF). Each of these indices has been proposed for the prediction of functional recovery. Their rel... Abstract

American journal of physiology. Heart and circulatory physiology, Volume 282, Issue 4, 1 1 2002, Pages H1350-8 Indexes of diastolic RV function: load dependence and changes after chronic RV pressure overload in lambs. Leeuwenburgh BP, Steendijk P, Helbing WA, Baan J
Diastolic function is a major determinant of ventricular performance, especially when loading conditions are altered. We evaluated biventricular diastolic function in lambs and studied possible load dependence of diastolic parameters [minimum first derivative of pressure vs. time (dP/dt(min)) and time constant of isovolumic relaxation (tau)] in normal (n = 5) and chronic right ventricular (RV) pressure-overloaded (n = 5) hearts by using an adjustable band on the pulmonary artery (PAB). Pressure-... Abstract

British journal of cancer, Volume 86, Issue 5, 1 1 2002, Pages 811-818 A fully human anti-Ep-CAM scFv-beta-glucuronidase fusion protein for selective chemotherapy with a glucuronide prodrug. de Graaf M, Boven E, Oosterhoff D, van der Meulen-Muileman IH, Huls GA, Gerritsen WR, Haisma HJ, Pinedo HM
Monoclonal antibodies against tumour-associated antigens could be useful to deliver enzymes selectively to the site of a tumour for activation of a non-toxic prodrug. A completely human fusion protein may be advantageous for repeated administration, as host immune responses may be avoided. We have constructed a fusion protein consisting of a human single chain Fv antibody, C28, against the epithelial cell adhesion molecule and the human enzyme beta-glucuronidase. The sequences encoding C28 and h... Abstract

European journal of nuclear medicine and molecular imaging, Volume 29, Issue 4, 19 3 2002, Pages 452-457 Feasibility, safety and image quality of cardiac FDG studies during hyperinsulinaemic-euglycaemic clamping. Bax JJ, Visser FC, Poldermans D, van Lingen A, Elhendy A, Boersma E, Visser CA
Fluorine-18 fluorodeoxyglucose (FDG) imaging for the assessment of myocardial viability has become an integral part of the diagnostic and prognostic work-up of patients with ischaemic cardiomyopathy. To ensure good image quality, in particular in patients with diabetes mellitus, hyperinsulinaemic-euglycaemic clamping has been proposed. In this study we evaluated the safety and the image quality of cardiac FDG imaging during clamping in a large group of patients, including a subgroup with diabete... Abstract

Netherlands heart journal : monthly journal of the Netherlands Society of Cardiology and the Netherlands Heart Foundation, Volume 10, Issue 2, 1 1 2002, Pages 43-47 Rhythmic sensory stimulation improves fitness by conditioning the autonomic nervous system. Frederiks J, Swenne CA, Ghafoerkhan A, Lalmahomed H, Maan AC, Schalij MJ, Bruschke AV, van der Wall EE
Background: Endurance training is known to alter the functioning of the autonomic nervous system, a major goal when pursuing fitness. Here, we test the hypothesis that the training-associated rhythmic sensations alone, hence without the usual accompanying physical exercise, accomplish this effect. Method: We studied sixteen resting healthy male volunteers, age (mean±SD) 25.9±3.7 years. During one hour we applied, at marching pace (2 bursts per second), bipolar transcutaneous electrical sensory n... Abstract

British journal of cancer, Volume 86, Issue 3, 1 1 2002, Pages 436-442 Prerequisites for effective adenovirus mediated gene therapy of colorectal liver metastases in the rat using an intracellular neutralizing antibody fragment to p21-Ras. van Etten B, ten Hagen TL, de Vries MR, Ambagtsheer G, Huet T, Eggermont AM
Ras mutations are present in 40-50% of colorectal cancers. Inactivating this oncogene may therefore reduce proliferation capacity. In order to target ras we studied the transduction efficacy and anti tumour activity of an adenoviral vector expressing an intracellular, neutralizing single chain antibody to p21-ras (Y28). In in vitro studies transfection levels of the K-ras mutated rat colon carcinoma cell line CC531 were studied using the LacZ marker gene. In our in vivo liver metastases model di... Abstract

Clinical genetics, Volume 61, Issue 2, 1 1 2002, Pages 115-125 ABCA1 regulatory variants influence coronary artery disease independent of effects on plasma lipid levels. Zwarts KY, Clee SM, Zwinderman AH, Engert JC, Singaraja R, Loubser O, James E, Roomp K, Hudson TJ, Jukema JW, Kastelein JJ, Hayden MR
The authors have previously shown that individuals heterozygous for ABCA1 mutations have decreased high density lipoprotein cholesterol, increased triglycerides and an increased frequency of coronary artery disease (CAD), and that single nucleotide polymorphisms (SNPs) in the coding region of the ABCA1 gene significantly impact plasma lipid levels and the severity of CAD in the general population. They have now identified several SNPs in non-coding regions of ABCA1 which may be important for the... Abstract
