Annals of vascular surgery, Volume 21, Issue 6, 1 1 2007, Pages 780-789 Glycemic control, lipid-lowering treatment, and prognosis in diabetic patients with peripheral atherosclerotic disease. Feringa HH, Karagiannis SE, Vidakovic R, Elhendy A, Schouten O, Boersma E, Bax JJ, Poldermans D
Glycemic control may be an underestimated risk factor in diabetic patients with peripheral arterial disease (PAD). Chronic statin therapy may improve glycemic control and outcome in these patients. In an observational cohort study of 425 consecutive diabetic patients with PAD, chronic statin therapy was noted, the ankle-brachial index was measured, and serial glycemic hemoglobin (HbA(1c)) measurements were obtained. During follow-up (median 7 years), all-cause mortality and cardiac death occurre... Abstract

Stem cell research, Volume 1, Issue 2, 1 1 2007, Pages 129-137 Reduction of myocardial infarct size by human mesenchymal stem cell conditioned medium. Timmers L, Lim SK, Arslan F, Armstrong JS, Hoefer IE, Doevendans PA, Piek JJ, El Oakley RM, Choo A, Lee CN, Pasterkamp G, de Kleijn DP
Although paracrine effects of mesenchymal stem cells (MSCs) have been suggested previously, cardioprotection by human MSC secretions has never been demonstrated. Human MSC-conditioned medium (CM) was collected by following a clinically compliant protocol. In a porcine model of ischemia and reperfusion injury, intravenous and intracoronary MSC-CM treatment significantly reduced myocardial nuclear oxidative stress as determined by immunostaining for 8-hydroxy-2'-deoxyguanosine. In addition, e... Abstract

The left atrium (LA) anterior-posterior diameter was one of the first standardised echocardiographic parameters. However, the clinical importance of LA size assessment has been neglected for some time. Recent population-based studies have demonstrated the prognostic value of LA size for long-term outcome. Furthermore, with new dedicated techniques such as tissue Doppler imaging, it has become feasible to assess (regional) LA function. In addition, the introduction of catheter ablation procedures... Heart (British Cardiac Society), Volume 93, Issue 11, 1 1 2007, Pages 1339-1350 Optimal use of echocardiography in cardiac resynchronisation therapy. Bleeker GB, Yu CM, Nihoyannopoulos P, de Sutter J, Van de Veire N, Holman ER, Schalij MJ, van der Wall EE, Bax JJ
Echocardiography has several roles in patients with cardiac resynchronisation therapy (CRT). First, it can optimise selection of CRT candidates by demonstration of left ventricular (LV) dyssynchrony. Second, it can be used to assess immediate response to CRT, including detection of acute LV resynchronisation. Echocardiography is also useful to evaluate long-term benefit from CRT. Finally, echocardiography is important in optimisation of pacemaker settings, including AV and VV optimisation. Abstract

Circulation, Volume 116, Issue 20, 29 5 2007, Pages 2241-2252 Catheter ablation of ventricular tachycardia after repair of congenital heart disease: electroanatomic identification of the critical right ventricular isthmus. Zeppenfeld K, Schalij MJ, Bartelings MM, Tedrow UB, Koplan BA, Soejima K, Stevenson WG
Background: Catheter ablation of ventricular tachycardia (VT) after repair of congenital heart disease can be difficult because of nonmappable VTs and complex anatomy. Insights into the relation between anatomic isthmuses identified by delineating unexcitable tissue using substrate mapping techniques and critical reentry circuit isthmuses might facilitate ablation. Methods and results: Sinus rhythm voltage mapping of the right ventricle was performed in 11 patients with sustained VT after repair... Abstract

Pathology, research and practice, Volume 203, Issue 12, 29 5 2007, Pages 863-872 Activation of signaling molecules and matrix metalloproteinases in right ventricular myocardium of rats with pulmonary hypertension. Umar S, Hessel M, Steendijk P, Bax W, Schutte C, Schalij M, van der Wall E, Atsma D, van der Laarse A
Pulmonary hypertension induces right ventricular (RV) overload, which is transmitted to cardiomyocytes via integrins that activate intracellular messengers, including focal adhesion kinase (FAK) and neuronal nitric oxide synthase (NOS1). We investigated whether RV hypertrophy (RVH) and RV failure (RVF) were associated with activation of FAK, NOS1, and matrix metalloproteinases (MMPs). Rats were treated without (RVC) or with a low dose of monocrotaline (30mg/kg) to induce RVH, and with a high dos... Abstract

The American journal of cardiology, Volume 100, Issue 12, 26 4 2007, Pages 1786-1791 Comparison with computed tomography of two ultrasound devices for diagnosis of abdominal aortic aneurysm. Vidakovic R, Feringa HH, Kuiper RJ, Karagiannis SE, Schouten O, Dunkelgrun M, Hoeks SE, Bom N, Bax JJ, Neskovic AN, Poldermans D
Screening for abdominal aortic aneurysms (AAAs) in patients at risk will become more cost effective if a simple, inexpensive, and reliable ultrasound device is available. The aim of this study was to compare a 2-dimensional, handheld ultrasound device and a newly developed ultrasound volume scanner (based on bladder scan technology) with computed tomography (CT) for diagnosing AAA. A total of 146 patients (mean age 69 +/- 10 years; 127 men) were screened for the presence of AAAs (diameter >3... Abstract

The American journal of cardiology, Volume 100, Issue 11, 24 4 2007, Pages 1665-1670 Usefulness of QRS duration to predict response to cardiac resynchronization therapy in patients with end-stage heart failure. Mollema SA, Bleeker GB, van der Wall EE, Schalij MJ, Bax JJ
Despite current selection criteria, 20% to 30% of patients treated with cardiac resynchronization therapy (CRT) do not benefit. It has been suggested that QRS duration may not be the optimal criterion to select patients for CRT. The objective of this study was to systematically evaluate the predictive value of QRS duration for response to CRT in a large group of consecutive patients. A total of 242 patients with heart failure scheduled for implantation of a CRT device were studied. Selection cri... Abstract

Green fluorescent proteins (GFPs) and variants thereof are widely used to study protein localization and dynamics. We engineered a specific binder for fluorescent proteins based on a 13-kDa GFP binding fragment derived from a llama single chain antibody. This GFP-binding protein (GBP) can easily be produced in bacteria and coupled to a monovalent matrix. The GBP allows a fast and efficient (one-step) isolation of GFP fusion proteins and their interacting factors for biochemical analyses includin... Abstract

Background: Nonresponse to cardiac resynchronization therapy is associated with the presence of slow or nonconducting scar tissue. Genetic modification of scar tissue, aimed at improving conduction, may be a novel approach to achieve effective resynchronization. Therefore, the feasibility of resynchronization with genetically modified human ventricular scar fibroblasts was studied in a coculture model. Methods and results: An in vitro model was used to study the effects of forced expression of t... Abstract

Pflugers Archiv : European journal of physiology, Volume 455, Issue 6, 2 1 2007, Pages 979-986 Release of cardiac troponin I from viable cardiomyocytes is mediated by integrin stimulation. Hessel MH, Atsma DE, van der Valk EJ, Bax WH, Schalij MJ, van der Laarse A
Elevated cardiac troponin-I (cTnI) levels have been demonstrated in serum of patients without acute coronary syndromes, potentially via a stretch-related process. We hypothesize that this cTnI release from viable cardiomyocytes is mediated by stimulation of stretch-responsive integrins. Cultured cardiomyocytes were treated with (1) Gly-Arg-Gly-Asp-Ser (GRGDS, n = 22) to stimulate integrins, (2) Ser-Asp-Gly-Arg-Gly (SDGRG, n = 8) that does not stimulate integrins, or (3) phosphate-buffered saline...
