British journal of cancer, Volume 86, Issue 5, 1 1 2002, Pages 811-818 A fully human anti-Ep-CAM scFv-beta-glucuronidase fusion protein for selective chemotherapy with a glucuronide prodrug. de Graaf M, Boven E, Oosterhoff D, van der Meulen-Muileman IH, Huls GA, Gerritsen WR, Haisma HJ, Pinedo HM
Monoclonal antibodies against tumour-associated antigens could be useful to deliver enzymes selectively to the site of a tumour for activation of a non-toxic prodrug. A completely human fusion protein may be advantageous for repeated administration, as host immune responses may be avoided. We have constructed a fusion protein consisting of a human single chain Fv antibody, C28, against the epithelial cell adhesion molecule and the human enzyme beta-glucuronidase. The sequences encoding C28 and h... Abstract

European journal of nuclear medicine and molecular imaging, Volume 29, Issue 4, 19 3 2002, Pages 452-457 Feasibility, safety and image quality of cardiac FDG studies during hyperinsulinaemic-euglycaemic clamping. Bax JJ, Visser FC, Poldermans D, van Lingen A, Elhendy A, Boersma E, Visser CA
Fluorine-18 fluorodeoxyglucose (FDG) imaging for the assessment of myocardial viability has become an integral part of the diagnostic and prognostic work-up of patients with ischaemic cardiomyopathy. To ensure good image quality, in particular in patients with diabetes mellitus, hyperinsulinaemic-euglycaemic clamping has been proposed. In this study we evaluated the safety and the image quality of cardiac FDG imaging during clamping in a large group of patients, including a subgroup with diabete... Abstract

Netherlands heart journal : monthly journal of the Netherlands Society of Cardiology and the Netherlands Heart Foundation, Volume 10, Issue 2, 1 1 2002, Pages 43-47 Rhythmic sensory stimulation improves fitness by conditioning the autonomic nervous system. Frederiks J, Swenne CA, Ghafoerkhan A, Lalmahomed H, Maan AC, Schalij MJ, Bruschke AV, van der Wall EE
Background: Endurance training is known to alter the functioning of the autonomic nervous system, a major goal when pursuing fitness. Here, we test the hypothesis that the training-associated rhythmic sensations alone, hence without the usual accompanying physical exercise, accomplish this effect. Method: We studied sixteen resting healthy male volunteers, age (mean±SD) 25.9±3.7 years. During one hour we applied, at marching pace (2 bursts per second), bipolar transcutaneous electrical sensory n... Abstract

British journal of cancer, Volume 86, Issue 3, 1 1 2002, Pages 436-442 Prerequisites for effective adenovirus mediated gene therapy of colorectal liver metastases in the rat using an intracellular neutralizing antibody fragment to p21-Ras. van Etten B, ten Hagen TL, de Vries MR, Ambagtsheer G, Huet T, Eggermont AM
Ras mutations are present in 40-50% of colorectal cancers. Inactivating this oncogene may therefore reduce proliferation capacity. In order to target ras we studied the transduction efficacy and anti tumour activity of an adenoviral vector expressing an intracellular, neutralizing single chain antibody to p21-ras (Y28). In in vitro studies transfection levels of the K-ras mutated rat colon carcinoma cell line CC531 were studied using the LacZ marker gene. In our in vivo liver metastases model di... Abstract

Circulation, Volume 105, Issue 6, 1 1 2002, Pages 702-706 Cardiac autonomic dysfunction in Brugada syndrome. Wichter T, Matheja P, Eckardt L, Kies P, Schäfers K, Schulze-Bahr E, Haverkamp W, Borggrefe M, Schober O, Breithardt G, Schäfers M
Background: Patients with Brugada syndrome present with characteristic ECG abnormalities (atypical right bundle-branch block and ST-segment elevation) and life-threatening ventricular tachyarrhythmias despite structurally normal hearts. Involvement of the autonomic nervous system is suggested by the occurrence of ventricular tachyarrhythmias and sudden death at rest or during sleep and by changes of typical ECG signs under pharmacological modulation of the myocardial autonomic tone. Methods and... Abstract

Clinical genetics, Volume 61, Issue 2, 1 1 2002, Pages 115-125 ABCA1 regulatory variants influence coronary artery disease independent of effects on plasma lipid levels. Zwarts KY, Clee SM, Zwinderman AH, Engert JC, Singaraja R, Loubser O, James E, Roomp K, Hudson TJ, Jukema JW, Kastelein JJ, Hayden MR
The authors have previously shown that individuals heterozygous for ABCA1 mutations have decreased high density lipoprotein cholesterol, increased triglycerides and an increased frequency of coronary artery disease (CAD), and that single nucleotide polymorphisms (SNPs) in the coding region of the ABCA1 gene significantly impact plasma lipid levels and the severity of CAD in the general population. They have now identified several SNPs in non-coding regions of ABCA1 which may be important for the... Abstract

A 75-year-old man complaining of acute abdominal pain, 1 month after elective surgical repair of an abdominal aortic aneurysm by an aortabi-iliac bypass graft, was referred and admitted to the emergency room. Imaging by sonography and computed tomography scan revealed a ruptured iliac pseudoaneurysm at the right iliac anastomotic site with associated large retroperitoneal hematoma. We inserted a self-expanding covered Z-stent graft by a transfemoral approach and the iliac anastomotic pseudoaneur... Abstract

The selective activation of a relatively non-toxic prodrug by an enzyme present only in the tumour should enhance the drug concentration at the tumour site and result in a better anti-tumour effect and a reduction in systemic toxicity as compared to conventional chemotherapy. beta-Glucuronidase is such an enzyme. It is normally expressed in the lysosomes of cells. In larger tumours, however, high levels of the enzyme are present in necrotic areas. Several glucuronide prodrugs have been synthesis... Abstract

Journal of cardiovascular pharmacology, Volume 39, Issue 1, 1 1 2002, Pages 61-72 Calcium sensitizer EMD 57033, but not the beta1-adrenoreceptor agonist dobutamine, increases mechanical efficiency in stunned myocardium. Trines SA, Smits CA, van der Moer J, Slager CJ, Verdouw PD, Krams R
External work, efficiency of energy transfer (EET), and mechanical efficiency (defined as the ratio of external work over myocardial oxygen consumption (MVO2) are reduced in stunned myocardium. We therefore evaluated how inotropic stimulation by dobutamine and the calcium sensitizer EMD 57033 affected the regional stress-strain relationship as, from which contractility (E(es)), external work at the working point (EWwp), maximal external work (EWmax), EETwp (%), and EETmax are determined. Journal of cardiovascular magnetic resonance : official journal of the Society for Cardiovascular Magnetic Resonance, Volume 4, Issue 3, 1 1 2002, Pages 353-357 Noninvasive and invasive evaluation of noncompaction cardiomyopathy. Bax JJ, Atsma DE, Lamb HJ, Rebergen SA, Bootsma M, Voogd PJ, Somer S, Schalij MJ, de Roos A, van der Wall EE
Noncompaction cardiomyopathy is a recently described rare congenital cardiomyopathy; patients can be asymptomatic or develop diastolic and/or systolic left ventricular dysfunction with heart failure, systemic emboli or ventricular arrhythmias. Long-term prognosis is poor. Currently, diagnosis is based on findings on 2D echocardiography; in the current case report we demonstrate the use of MRI to diagnose noncompaction cardiomyopathy.
