Therapeutic drug monitoring, Volume 40, Issue 5, 1 1 2018, Pages 515-525 Liquid Biopsies to Monitor Solid Organ Transplant Function: A Review of New Biomarkers. Verhoeven JGHP, Boer K, Van Schaik RHN, Manintveld OC, Huibers MMH, Baan CC, Hesselink DA
Despite modern immunosuppressive therapy, allograft rejection remains a major cause of solid organ transplant dysfunction. For clinical care, organ transplant function is routinely monitored by measuring biomarkers that, depending on the organ transplanted, include serum creatinine, N-terminal pro-hormone of brain natriuretic peptide (NT-proBNP), and aspartate aminotransferase. All can be measured easily in clinical chemistry laboratories. The main problem with these biomarkers is that they have... Abstract

Since its clinical introduction in 1998, the topoisomerase I inhibitor irinotecan has been widely used in the treatment of solid tumors, including colorectal, pancreatic, and lung cancer. Irinotecan therapy is characterized by several dose-limiting toxicities and large interindividual pharmacokinetic variability. Irinotecan has a highly complex metabolism, including hydrolyzation by carboxylesterases to its active metabolite SN-38, which is 100- to 1000-fold more active compared with irinotecan... Abstract

Environmental science & technology, Volume 52, Issue 20, 27 4 2018, Pages 11500-11509 Patterns of Host-Associated Fecal Indicators Driven by Hydrology, Precipitation, and Land Use Attributes in Great Lakes Watersheds. Dila DK, Corsi SR, Lenaker PL, Baldwin AK, Bootsma MJ, McLellan SL
Fecal contamination from sewage and agricultural runoff is a pervasive problem in Great Lakes watersheds. Most work examining fecal pollution loads relies on discrete samples of fecal indicators and modeling land use. In this study, we made empirical measurements of human and ruminant-associated fecal indicator bacteria and combined these with hydrological measurements in eight watersheds ranging from predominantly forested to highly urbanized. Flow composited river samples were collected over l... Abstract

eLife, Volume 7, 27 4 2018, Pages e41076 Optogenetics enables real-time spatiotemporal control over spiral wave dynamics in an excitable cardiac system. Majumder R, Feola I, Teplenin AS, de Vries AA, Panfilov AV, Pijnappels DA
Propagation of non-linear waves is key to the functioning of diverse biological systems. Such waves can organize into spirals, rotating around a core, whose properties determine the overall wave dynamics. Theoretically, manipulation of a spiral wave core should lead to full spatiotemporal control over its dynamics. However, this theory lacks supportive evidence (even at a conceptual level), making it thus a long-standing hypothesis. Here, we propose a new phenomenological concept that involves a... Abstract

The volume of the lateral ventricles (LV) increases with age and their abnormal enlargement is a key feature of several neurological and psychiatric diseases. Although lateral ventricular volume is heritable, a comprehensive investigation of its genetic determinants is lacking. In this meta-analysis of genome-wide association studies of 23,533 healthy middle-aged to elderly individuals from 26 population-based cohorts, we identify 7 genetic loci associated with LV volume. These loci map to chrom... Abstract

The American journal of cardiology, Volume 123, Issue 1, 26 4 2018, Pages 75-83 Effect of Functional Mitral Regurgitation on Outcome in Patients Receiving Cardiac Resynchronization Therapy for Heart Failure. van der Bijl P, Khidir M, Ajmone Marsan N, Delgado V, Leon MB, Stone GW, Bax JJ
Functional mitral regurgitation (FMR) is common in heart failure (HF), and negatively impacts prognosis. Cardiac resynchronization therapy (CRT) can improve FMR, but the long-term changes in and impact of FMR after CRT are still unclear. The present study investigated the prevalence, evolution and impact on mortality of FMR before and after CRT in patients with HF. A total of 1,313 patients (66 ± 11 years, 77% male, 59% ischemic heart disease) treated with CRT were evaluated. Patients were divid... Abstract

Healthcare (Basel, Switzerland), Volume 6, Issue 3, 19 3 2018, Pages E118 The Applicability of N: Ancient Debates and Modern Experimental Design. Van Schaik KD
Medicine has always been characterized by a tension between the particular and the general. A clinician is obligated to treat the individual in front of her, yet she accomplishes this task by applying generalized knowledge that describes an abstract average but not necessarily a specific person. Efforts to systematize this process of moving between the particular and the general have led to the development of randomized controlled trials and large observational studies. Inclusion of tens of thou... Abstract

European journal of heart failure, Volume 20, Issue 11, 17 3 2018, Pages 1601-1610 Rationale and design of the phase 2b clinical trials to study the effects of the partial adenosine A1-receptor agonist neladenoson bialanate in patients with chronic heart failure with reduced (PANTHEON) and preserved (PANACHE) ejection fraction. Voors AA, Shah SJ, Bax JJ, Butler J, Gheorghiade M, Hernandez AF, Kitzman DW, McMurray JJV, Wirtz AB, Lanius V, van der Laan M, Solomon SD
Despite major advances in the treatment of chronic heart failure (HF) with reduced ejection fraction (HFrEF), morbidity and mortality associated with the condition remain high, suggesting the need for additional treatment options, particularly haemodynamically neutral treatments that do not alter blood pressure, heart rate, or renal function. HF with preserved ejection fraction (HFpEF) is also associated with high morbidity and mortality and adequate treatment options are limited; thus there is... Abstract

Aquatic animals live in an acoustic world in which they often rely on sound detection and recognition for various aspects of life that may affect survival and reproduction. Human exploitation of marine resources leads to increasing amounts of anthropogenic sound underwater, which may affect marine life negatively. Marine mammals and fishes are known to use sounds and to be affected by anthropogenic noise, but relatively little is known about invertebrates such as decapod crustaceans. We conducte... Abstract

Pharmacogenomic (PGx) testing is gaining recognition from physicians, pharmacists and patients as a tool for evidence-based medication management. However, seemingly similar PGx testing panels (and PGx-based decision support tools) can diverge in their technological specifications, as well as the genetic factors that determine test specificity and sensitivity, and hence offer different values for users. Reluctance to embrace PGx testing is often the result of unfamiliarity with PGx technology, a... Abstract

Background: Plaque angiogenesis is associated with atherosclerotic lesion growth, plaque instability and negative clinical outcome. Plaque angiogenesis is a natural occurring process to fulfil the increasing demand of oxygen and nourishment of the vessel wall. However, inadequate formed, immature plaque neovessels are leaky and cause intraplaque haemorrhage. Objective: Blockade of VEGFR2 normalizes the unbridled process of plaque neovessel formation and induces maturation of nascent vessels resu... Abstract
